▲ [20p-C205-6] [INVITED] Wavefront Engineered Light-Needle Microscopy Utilizing Multiplexed Computer-Generated Holograms

We propose a novel imaging technique that can retrieve the axial information in light needle scanning fluorescence microscopy by introducing a method based on a multiplexed computer-generated hologram applied to fluorescence signals. By this technique, axially resolved three-dimensional images can be obtained from a single two-dimensional raster scanning of a needle spot, which remarkably increases the acquisition speed. We demonstrate real-time and video-rate acquisitions of three-dimensional images for fluorescence specimens. The prompt observation of thicker biological samples is also achieved using our method.
