An Approach to 3D Super-Resolution by Localization of Single Particles

We present a technique to overcome conventional diffraction-limited resolution in the in-plane direction to realize nearly isotropic nano-resolution with the use of an ultrathin X-ray light sheet microscope. Automated processing with stochastic optical reconstruction microscopy (STORM) and photo-activated localization microscopy (PALM) are used to create a three-dimensional map of the localization of the excited source, which is used to derive a promising method to discern single particles from multiple particle aggregations.
