[18a-E302-9] Evaluation of Diffusion of Lipid Molecules Using Quenching by Co2+ Ions

Previously, we have studied the lateral diffusion of supported lipid bilayer membranes using fluorescence recovery after photobleaching technique, and have reported that the diffusion consists of two components when positively charged Poly(L-ornithine) is coated on the substrate surface. In this study, we attempted to quench the leaflets on one side of the lipid bilayer using Co2+ ions. Assuming that it consists of two components, the lower-leaflet, which is close to the substrate surface and interacts strongly with the substrate, and the upper-leaflet, which is far from the substrate surface and is not easily affected by surface charges. Diffusion in each leaflet has been separated using the quenching effect of Co2+ ions and evaluated.
