[ODP-078] De novo assembly of Actinomyces capricornis strain MAS-1T complete genome sequence
We report complete genome sequence of Actinomyces capricornis strain MAS-1T, which was isolated from Japanese serow oral cavity. The complete genome of strain MAS-1T was determined using single-molecule real-time technology (SMRT). A library containing 20 kbp inserts was created using SMRTbell and analyzed with PacBio Sequel system. De novo assembly of the multiplexed microbial genome data generated by the PacBio RS II sequencer was performed using the SMRT_link. Digital DNA-DNA hybridization (dDDH) and average nucleotide identity (ANI) between MAS-1T and its related species were performed using Genome to Genome Distance Calculator (http://ggdc.dsmz.de) and ANI calculator (http://enve-omics.ce.gatech.edu/ani/index), respectively. The complete genome of strain MAS-1T consisted of 3,170,963 bp in a single circular chromosome with a G+C content of 71.3%. Annotation using DFAST predicted 2,606 CDSs, 9 rRNAs, 59 tRNAs, and 4 CRISPRS. The dDDH and ANI values between strain MAS-1T and its related species were 23.5-39.5% and 82.11-91.01%, respectively, which were below the threshold (70% and 95%, respectively) for species delineation, indicating that strain MAS-1TT represents a novel species. The complete genome sequence of strain MAS-1T has been deposited in DDBJ under accession number AP025017.
