[O3-E-PM1-01] Resvertrol downregulates Fusobacterium nucleatum-induced epithelial-mesenchymal transition in oral cancer cells

During cancer development, epithelial-mesenchymal transition (EMT) plays an important role in promoting tumor cell invasion and migration. Fusobacterium nucleatum (F. nucleatum), an anaerobic opportunistic pathogen associated with periodontitis, has been reported to be associated with the pathogenesis of oral squamous cell carcinoma. Resveratrol is an essential polyphenol for plants and exhibits anti-inflammatory and anticancer properties. The objective of this study was to examine the effect of resveratrol on the tumor-promoting activity of F. nucleatum. We evaluated the proliferation and migration ability of HSC-3 cells using the Cell Counting Kit-8 and wound healing assay. And under the microscope, HSC-3 cells co-cultured with F.nucleatum have changed from a paving stone, sheet-like structure to a fibroblast-like spindle shape. The expression of EMT transcription factor snai1 was enhanced, while E-cadherin was decreased in Real-time RT-PCR and Western blotting, indicating that F. nucleatum induces migratory ability in HSC-3 cells. On the other hand, resveratrol suppressed these expressions by F. nucleatum in HSC-3 cells. These results suggest that resveratrol regulates F. nucleatum-induced cancer cell migration in vitro and that the regulatory mechanism may be mediated by the snai1 signaling pathway.
