[13p-PB7-14] Development of SERS substrates by seed-mediated growth of silver on gold-nanoparticles for biosensing applications

Surface-enhanced Raman spectroscopy (SERS) is a technique that utilizes noble metal nanostructures to enhance Raman scattering of adsorbing molecules. SERS has a good sensitivity, selectivity, and fingerprint characteristics. Noble metal nanostructures with reproducible and large enhancement of SERS signal are required for applications to the label-free biosensing. In this research, we study on the fabrication of SERS substrate by using seed-mediated growth of Ag nanostructures on Au nanoparticles.

Au nanoparticles were densely decorated on a glass substrate by sputtering-annealing method. Ag was deposited on the Au nanoparticles by processing the substrate with a growth solution including Ag+ ion. The color of substrate changed from pink to dark green due to the plasmon resonance of Au and Ag nanoparticles. 4-aminothiophenol (4-ATP:100μM) and bilirubin (100μM) was dropped onto the substrate as target molecules. SERS spectra were measured by focusing a green laser beam on the substrate under an optical microscope. Clear SERS peaks of 4-ATP and bilirubin were observed, indicating that this substrate can be used for label-free bio-sensing application.
