Live cell imaging is an excellent technique to visualize the dynamics and activities of tissue, cells, and individual organelles in living cells. In order to evaluate toxicity of drugs and foods to living cells by time-lapse observation of labeled subcellular structures, a wide variety of fluorescent proteins, such as green fluorescent protein (GFP) and cyan fluorescent protein (CFP), are isolated and introduced into specific sites in living cells. In those researches, the photodamage which is induced by the light illumination for excitation of fluorescent proteins becomes an important issue for the accurate assessment of the toxicity. In this study, we quantitatively investigate the phototoxicity of short-wavelength visible excitation lights (blue and green) to MDA-435 living cells in which chromatin is simultaneously labeled with blue (ECFP) and red (Plum) fluorescent proteins.
