[16p-2C-3] Multiple single-molecule tracking in nanoscale based on spectral discrimination

Simultaneous nanometric tracking of multiple proteins has been realized by a simple combination of multicolor fluorescent labeling of target proteins and imaging spectroscopy. This method can observe dynamic behaviors of multiple molecules within sub-diffraction-limit scale. Using quantum dot probes of distinct colors, we experimentally verified that our scheme provides a localization precision of a few nanometers at the video rate. One-dimensional processive movement of two heads of a single myosin molecule and multiple myosin molecules was successfully traced. Furthermore, we modified the microscopy system for two-dimensional measurement and applied it to tracking of multiple myosin molecules. The presented scheme would be useful for investigating cooperative movement of proteins in supramolecular nanomachinery.
