[14p-P18-10] Characterization of Protein Aggregation and Crystallization by in-situ Forward Light Scattering Measurement

Keywords:aggregate, light scattering, protein

We have developed an apparatus which can perform in-situ measurements of forward-light scattering sensitive to protein aggregates. Characterization of lysozyme (HEWL) aggregation was evaluated for solutions near a quasi-stabilization area, using the apparatus. HEWL solutions at high salt concentration showed the characteristic pattern that became a function of negative non-integer power. This means a formation of dense fractal aggregates, which results in generating crystal growth. Addition of lower salt produced HEWL aggregates with a coarse structure, which did not drive crystal growth.
