We have examined in-situ the viscoelasticity change of stratum corneum and corneocytes in aqueous solutions by the force curve measurement using an AFM. However, the measurement errors of Young’s modulus included one order of magnitude for different locations. To solve this problem, we put the corneocytes on gel seat, which is often used in such as cell culture, so that a longer loading path of the AFM tip can be attained reducing the measurement error. In this study, we prepared the various stiffness of gel seat and examined the force curve of corneocyte on the gels.
