[15a-B8-11] Optimization of continuous flow PCR for highly specific pre-amplification of BEAMing

Recently, BEAMing (Beads Emusion Amplification and Magnetics) is one of detection technology for the high sensitive genetic mutation. BEAMing is attracted attention in term of detection for circulating tumor DNA in the blood. This method has 7 steps (pre-amplification/ dilution/ emulsification/ emulsion-PCR/ emulsion break/ hybridization/ FCM) and these steps is carried out by manual method. Manual based BEAMing method is complicated and takes many times. Therefore, we are trying to realize automatic and rapid BEAMing method by using microfluidic devices. In this study, we investigated the specificity and uniformity of pre-amplification process and report on the result of the verification of the applicability to BEAMing.
