[20p-P12-14] Selection and Characterization of 3-(1-naphoyl)indole derivatives-Specific Alpaca VHH antibodies using a Phage Display Library

A new alpaca VHH antibody library for 3-(1-naphtoyl)indole derivatives were developed from alpaca immunized with 7-(3-(1-naphthoyl)-1H-indol-1-yl)-heptanoic acid-key limpet hole protein conjugates (Hep-KLH) as the immunogen. The library size was 3× 107 for VHH fragments. Two 3-naphtoylindole derivatives -specific clones from the library were isolated using biopanning technology. The binding specificity of these clones was confirmed using a competitive enzyme-linked immunosorbent assay (c-ELISA). Based on the results of c-ELISA, a median inhibitory concentration (IC50) of purified two VHH antibodies exhibited an approximate 2×10-6 M and 3.2×10-5 M respectively.
