Carbohydrates as a kind of functional biomolecule play many critical roles in biological processes, such as cell-cell recognition, protein targeting, and nutrition. Develop a method which can analyze carbohydrates intracellular become more and more important for understanding their biological functions. Matrix-assisted laser desorption/ionization (MALDI) mass spectrometry is one of the analytical techniques has been extensively used for biomolecule analysis, due to its low sample consumption and high sensitivity. However, the conventional MALDI is suffering from poor ionization efficiency, poor shot-to-shot reproducibility and a serious matrix signal interference at the low mass region (<700 Da).In this study, a matrix-free carbohydrate analysis method was established by using the on-chip pulse-heating ionization method (PHI). Compared with conventional MALDI, a much clear mass spectra were obtained with very less background and fragmentation ion signals.
