[7p-A503-6] Formation enhancement of inter-digitated gel phase in DDAB lipid bilayer by DNA-functionalized nanoparticles

Previously, we developed a method for two-dimensional crystallization of DNA-functionalized nanoparticles (DNA-NPs). In our method, DNA-NPs adsorb onto lipid bilayer which is made by cationic lipid (DDAB) and diffuse on it via lipid molecules. Subsequently, DNA-NPs bind each other thorough DNA and two-dimensional crystal of DNA-NPs form. We have observed that DDAB lipid bilayer occurred phase separation and gel and interdigitated gel phases formed from liquid crystalline phase. Then, DNA-NPs preferably distributed into interdigitated gel phase. In order to reveal this cause, we compare the ratio of interdigitated gel phase in lipid bilayer and lipid bilayer with DNA-NPs. In lipid bilayer with DNA-NPs, formation of interdigitated gel phase was enhanced and ratio of interdigitated gel phase was twice as much as only lipid bilayer. This means that DNA-NPs do not distributed into interdigitated gel phase but lipid molecules which adsorb to DNA-NP preferably become interdigitated gel phase.
