[7p-S45-4] 3D Printing of Enhanced Green Fluorescent Protein
by Femtosecond Laser Cross-Linking Utilizing Rhodamine 6G

Microfabrication of enhanced green fluorescent protein (eGFP) nearby or possibly inside a living cell is expected to open a new door for study of cellular transport mechanisms. Two-photon polymerization technique using femtosecond (fs) laser direct writing (LDW) has been applied to fabricate 3D proteinaceous microstructures by photosensitized cross-linking of proteins. However, the developed technology is yet limited in the utilized types of proteins. Recently, we adopted this technique with 522 nm fs laser to 3D microfabrication of eGFP. Because eGFP is obtained by recombinant expression, its concentration is a restrictive aspect to microfabrication which can be counteracted by additional photosensitizers. Here, we report 3D fabrication utilizing Rhodamine 6G with a 522 nm fs laser. Based on absorption spectra of each constituent, one-photon absorption is probably initiating polymerization.
