Unlike the conventional matrix assisted laser desorption/ionization (MALDI) method which usually suffer from background signals, matrix-free techniques have become more and more popular for the analysis of low molecular weight (LMW) biological compounds. In the previous study, an on-chip pulse-heating ion source was established for simple inorganic sample analysis by only applying thermal energy from a single pulse without matrix, which indicate the ability of this ion source can be used for matrix-free protein analysis. This study describe the initial results of our efforts to obtain protein ion without matrix by the pulse-heating ion source. As a result, the single charged ion signal was obtained without matrix from Cytochrome C (MW: 12 kDa) and α-Lactalbumin (MW: 14.2 kDa), which are standards for protein MS analysis, respectively.
