Due to the heterogeneity within same cell population, a method for genome and transcriptome expression in single cell with positional information in tissues and cell network is demanded. Therefore, we are trying
to develop an analytical system using address-tag DNA, containing the positional information for the analyses of single cells. In this work, we are focusing on the combinatorial syntheses of address tag based oligonucleotides on chip using mild reagents compatible with PDMS polymer. A new deprotecting reagent was developed, and proved suitable for DNA synthesis using PDMS chip. We confirmed the sequence of synthesized oligonucleotide by MALDI-TOF/TOF mass spectrometry and gel electrophoresis.
