△ [18p-231C-3] Optimization of Focused Beams and Enhancement of Spatial Resolution of Subtraction Imaging using Vector Beams

The spatial resolution in confocal laser scanning microscopy (CLSM) is limited to about 200 nm because of the diffraction nature of light. Therefore, novel and simple methods that can overcome this limit are strongly desired. Our group has previously reported that the spatial resolution in CLSM is enhanced by subtraction imaging using vector beams without the appearance of serious negative side-lobes. In the present study, we present the results of a simulation study to find optimum combinations of vector beams in subtraction imaging to increase the signal intensity with reduced negative side-lobe generation while keeping the high spatial resolution.
