[20p-PA3-3] Quantification of Drug Side Effects on Cell-Free Synthesized hERG-Channels in Solvent-Free Lipid Bilayers Based on Microfabricated Silicon Chips

Ion channels, a type of membrane protein, have a gate mechanism for ion flow into and out of the cell membrane and have a function of selectively permeating ions. We aim to develop a drug screening system based on a reconstitution system in which ion channels are embedded in artificial lipid bilayer membrane, and this time we covered the hERG channel synthesized using cell-free synthesis system in this membrane system We tried to quantify drug side effects on this channel and compared it with the results of patch clamp method.
Micropores were prepared on a Si substrate by photolithography and wet etching, and a lipid bilayer membrane was formed in this micropore by a single molecule lamination method. Inhibitor was added, inhibition curves were generated based on the opening probability of hERG channel, and compared with patch clamp method.
