[17a-A202-3] Development of Single-Molecule Electrical Identification Method with Artificial Nucleotide Probe

e newly designed adenine-derivatives for improvement of the discrimination ability of single-molecule tunnel-current oligonucleotide sequencing. By using the oligonucleotide containing the synthesized nucleotide, we measured current-time profile by using our nano-gap electrode devices. Compared with adenosine, the conductances of the diaza-adenine derivatives are large so that the discrimination among other nucleotide is improved. This results suggest that artificial probe insertion for sample nucleotide would be one of promising methods for improvement of single-molecule tunnel-current oligonucleotide sequencing.
