We proposed a membrane damage assay using nonlinear optics as a new evaluation method of cytotoxicity, but the stability of the nonlinear optical signal was not good. The nonlinear signal tended to weaken as the cell shape was planar, and the damage was small when the number density of the cells was high. We improved the reproducibility of the membrane damage assay by reviewing the number of cells used for cell culture and the incubation time after the cell culture.
