Protein secretion monitoring provides vital information on intercellular activity that can be used for early indication of infectious diseases and regular healthcare. Current analytical methods include bulk cell observation, which can obscure crucial responses of individual cells within a large heterogeneous population. Therefore, in order to avoid overlooking such individual responses, it is necessary to isolate individual cells and observe their responses in real time, especially after perturbation with certain stimuli. To achieve this purpose, microwell structures, which are simple to fabricate yet effective in isolating and analyzing single cells, have been used. Integration of dielectrophoresis electrodes with microwells in the current study allowed for label free real-time analysis of protein signaling detected by the localized surface plasmon resonance (LSPR) observation method.
