[19p-A302-4] Superresolution nonlinear fluorescence microscopy using repetitive stimulated transition

We propose super-resolution nonlinear fluorescence microscopy with pump-probe setup that employs repetitive stimulated absorption and stimulated emission caused by two-color beams. The resulting nonlinear fluorescence that undergoes such a repetitive stimulated transition is detectable as a signal via the lock-in technique. The signal is produced by the multi-ply combination of incident beams, resulting in an improvement of the three-dimensional optical resolution. A phenomenological model of the nonlinear optical process, which is provided using rate equations, offers a new angle of interpretation of nonlinear fluorescence processes. The REST signal properties were experimentally confirmed to be in good agreement with the theoretical model. The proposed method is demonstrated as having superior optical resolution to that of conventional laser-scanning fluorescence microscopy.
