[18p-E204-1] Development of Axially Resolved, Three-Dimensional Light Needle Microscopy

We are developing a new method that can acquire a three-dimensional image from a single raster scanning of a light needle spot without moving the observation plane. We utilized a Bessel beam as a scanning laser spot (light needle spot) that has an extended focal depth elongated along the depth direction. Then, fluorescent signals are converted into Airy beams that exhibit self-bending property to retrieve the depth information of the samples. In this talk, we present the experimental demonstration of imaging for biological samples using our method. The applicability of the method is also discussed in detail.
