△ [19a-E203-1] Detection of α-Synuclein by liposome immobilized cantilever sensor using RT-QuIC method

We have detected α-synuclein (αSyn) recognized as causative agent for Parkinson Disease (PD) by phospholipid liposome-immobilized cantilever sensor. Its monomer existing usually in our brain cell starts to aggregate and grow as oligomers, protofibrils and fibrils. And these are found to be very toxic. it was revealed that aggregation of the monomeric αSyn is significantly promoted when a trace amount of αSyn fibril is mixed in the monomeric αSyn. Real-time quaking-induced conversion (RT-QuIC) method is based on the fibrilization phenomena. We focused on the RT-QuIC method. In this work, we have tried to apply RT-QuIC method to the cantilever sensor immobilizing phospholipid liposome for detecting a trace amount of αSyn aggregation at its initial stage.
