[21p-E206-3] Large-scale label-free single-cell analysis by vibrational flow cytometry

We present the principle of the recently developed high-throughput (1,000 cells/s) and broadband (> 1,000 cm-1) vibrational flow cytometer and its application to single-cell analysis of various cells. This method is enabled by the combination of time-domain Raman spectroscopy with femtosecond pulses and precise alignment cells in a microfluidic device with an acoustic standing wave. With this technique, we demonstrated large-scale and label-free quantification of proteins, carbohydrates, and lipids based on vibrational spectroscopic data obtained from every single cell.
