[10a-Z12-7] Single-Molecule Epigenetic Detection of micro RNA in colon cancer cell

We extracted colorectal cancer microRNAs and performed single molecules tunneling current for this microRNAs solution using nanogap electrodes. Using the characteristic conductance values of m6A and 5mC, the ratio of modified bases (m6A, and 5mC) in the obtained single-molecule signal was evaluated and determined. These values was found to be similar to those by mass spectrometry. Furthermore, the modification ratios of m6A and 5mC were evaluated for each of microRNA sequence number positions. As a result, it was found that there are differences depending on the sequence numbers, and we found that the ratio of methylation / non-methylation is closely related to the degree of agreement with the consensus sequence of the enzymes of m6A and 5mC.
