[10p-Z12-9] Development of the Plasmonic Sensor Chip for Quantitative Detection of Oral Candida in Human Saliva

Oral candida in saliva is known to be the important biomarker to measure host immunity. In the previous study, oral Candida bacteria was measured with plasmonic chip by grating-coupled surface plasmon-field enhanced fluorescence, and we developed a Candida assay to detection of Candida bacteria in human saliva samples and prepared the interface for sandwich assay on the chip. In this study, Candida bacteria in human saliva are detected quantitatively using the measurement system. The plasmonic chip was prepared by UV nano-imprint method and silver coating by rf-sputtering. The chip surface was coated with thin polydopamin film for absorbing protein G. After immobilization of anti-candida mannan IgG antibody, the heat-treated sample solution including Candida bacteria were injected. As a result of detection with a labeled fluorescence antibody, fluorescence intensities increased with a candida concentration, and oral candida bacteria in human saliva was quantitatively measured.
