[8a-Z12-5] Microfluidics of single cell interaction analysis for T cell activity evaluation

A specific T cell and antigen expressing APC interaction is essential in immune response. However, the interaction is very rare event due to the genomic rearrangement of the germline on TCR locus. It makes the analysis of single-cell-level pairing a challenge by using conventional methods. Thus, a new platform is required to promote, identify and single-cell-level measure T cell behavior in paring. This report describes that the results of the PDMS microfulidic device for single cell analysis of T cell-APC. Our device was developed for preparing the T cell-APC interaction and profiling each T cell reaction based on intracellular Ca2+ flux. This new platform to easily determine, retrieve and analyze the rare cell pair can advance the study about mechanism of immune system related T cells.
