The epithelial barrier, which composed of epithelial cells with tight junction, maintains homeostasis. On the other hand, it blocks drug absorption into the human body. Therefore, there is a high demand for carrier molecules that can deliver the biomolecule/drugs through the epithelial barrier in drug delivery systems. The energy-independent permeability of a phospholipid-mimicked amphiphilic random copolymer composed of 2-methacryloyloxyethyl phosphorylcholine (MPC) and n-butyl methacrylate (BMA) through cell membrane was discovered. Therefore, it can be expected as a carrier molecule. In this study, we evaluated its usefulness of phospholipid-mimicked amphiphilic random copolymer as an epithelial carrier by the ISFET based pH perturbation assay, which enables highly sensitive and selective evaluation of ion barrier cell membranes/tight junctions.
