The COVID-19 pandemic has created urgent demand for rapid detection of coronavirus (SARS-Cov-2). While PCR has been recognized as the most common way for the detection, it is still required to develop alternative techniques with higher detection sensitivity than conventional PCR. Surface plasmon resonance (SPR) biosensors are a promising alternative enabling label-free and real-time detection of antigen-antibody interactions in ultralow concentration. So far, there have been several attempts to improve the limit of detection (LOD) by depositing gold nanoparticles with diameter of several tens of nanometers onto the surface of gold thin films. Here, we revealed crucial plasmonic roles of relatively large nanoparticles with diameter of 150 nm in significant enhancement of the sensitivity, and detected SARS-Cov-2 nucleocapsid proteins at femtomolar level using the large gold nanoparticle-based SPR technique.
