The detection and analysis of exosomes, one of the endoplasmic reticulum secreted by cells, is thought to be useful for disease prediction because the microRNAs and proteins contained in them reflect the characteristics of the cells secreted. In our laboratory, we have shown that single exosomes of 100-200 nm in diameter can be detected by epi and transmission fluorescence microscopies using grating-coupled surface plasmon resonance (GC-SPR) enhanced fluorescence with a plasmonic chip coated with a silver thin film. In this study, quantitative evaluation of exosomes in solution was performed by transmission-type fluorescence microscopy and compared with the results obtained by epi-system.
