We demonstrate the world’s first four-wave mixing (FWM) and second-harmonic generation (SHG) imaging flow cytometry, enabling us to obtain a statistical number (N > 1,000) of single cells’ morphological information and to visualize non-centrosymmetric media in cells in a label-free manner. Our FWM and SHG imaging flow cytometer is composed of an FWM and SHG microscope with a 2D-rapid galvanometric scanner, a microfluidic device, and a high-speed image acquisition circuit, enabling a frame rate of 10 fps with a pixel resolution of 100 × 100 pixels. As a proof-of-principle demonstration, we used the FWM and SHG imaging flow cytometer to perform FWM and SHG imaging of polymer beads and Chromochloris zofingiensis (NIES-2175) cells. Our FWM and SHG flow cytometer paves the way for statistical studies of cellular dynamics and drug screening in a label-free manner.
