We are developing a new electrochemical impedance measurement method for real-time detection of the polymerase chain reaction (PCR). The basis of the method is the phenomenon that the synthesized DNA intercalator [Ru(bpy)2DPPZ]2+ promotes charge transfer between hexacyanoferrate ions and carbonaceous electrodes in PCR solution at a concentration of only 1 µM. It was experimentally confirmed that the effect is generated by the hexacyanoferrate ion, the [Ru(bpy)2DPPZ]2+ ion, and the carbonaceous electrode, and is independent of other ions such as electrolyte ions.
