A plasmonic chip, in which a wavelength-sized periodic structure is covered with a thin metal film, can provide an enhanced electric field by grating-coupled surface plasmon resonance (GC-SPR) and enhance the fluorescence. In our laboratory, highly sensitive fluorescence images of cells have been observed using a plasmonic chip. In this study, the fluorescence images of fluorescent particles (Dark Red) were observed with plasmonic chips having pitches of 430 and 480 nm under unpolarized and polarized excitation light in epi- and transmission optical systems. The plasmonic enhancement was discussed by excitation enhancement and fluorescence enhancement.
