[16a-Z21-4] Single molecule Electrical detection method Towards Comprehensive methylation-modified microRNA

MicroRNAs in urine were extracted and tunnel current single molecule measurement was performed using nanogap electrodes. Using the characteristic conductance of m6A as an index, the ratio of m6A-modified bases in the obtained monomolecular signal was evaluated, and the modification ratio of m6A in the sequence in the microRNA was determined. Furthermore, it was found that the modification rate of the specific sequence part of m6A changed between the cancer patient group and the healthy subject group. This value was similar to the previously reported value measured by mass spectrometry of microRNA. As a result, we were able to show the possibility of cancer diagnosis using the modification rate of m6A in specific microRNA as an index.
