Immune responses are elicited by T cells that specifically interact with antigen-presenting cells (APCs). T cell receptor (TCR) recognizes a peptide derived from a pathogen presented by APC and then T cell immunologically activates. However, the recognition is very rare event due to the genomic rearrangement of the germline on TCR locus. It makes the analysis of T cell activation which elicits immune responses a challenge by using conventional methods. Thus, a new platform is required to promote, detect and single-cell-level measure T cell behavior in paring. In this study, we developed a device for the formation of T cell-APC pairs at the single cell level and profiling each T cell activation based on intracellular Ca2+ flux as an indicator. In addition, we collected T cells after the measurement and analyzed by single cell RNA-Seq to link the intracellular Ca2+ signal and T cell gene expression.
