[17a-P06-2] Single exosome detection on a plasmonic chip under the transmitted-light fluorescence microscopy

Exosome is one of the extracellular vesicles with diameter of ~100 nm released from a cell. MicroRNAs and proteins are contained in exosomes, and therefore, studies of exosomes are known to be valuable for predicting diseases. In our laboratory, single exosomes have been detected by the epi-fluorescence microscopy using a plasmonic chip, which is a wavelength-sized periodic structure substrate coated with a thin metal film. Plasmonic chip can provide the enhanced fluorescence by the grating-coupled surface plasmon resonance (GC-SSPR) . In this study, we observed fluorescently labeled single exosomes with a plasmonic chip under the transmitted light, which is expected to improve a fluorescence enhancement compared with that of epi-fluorescence.
