▲ [18p-P02-4] Secondary Structure of Hair Protein Studied by Non-Negative Matrix Factorization and Raman Imaging

We applied Raman imaging and non-negative matrix factorization (NMF) into the cross sections of strands of gray human hair, specifically (i) the untreated hair (UN), (ii) the permanently treated curly hair (PE) and (iii) the permanently treated straight hair (ST), in order to reveal the microscopic mechanism of damages in hair through these perm treatments. We applied NMF into the amide I region of protein, giving reasonable and reproducible decompositions into the components corresponding to different secondary structures of protein and the scaling factors that developed images. This NMF approach indicated that the amount of α-helix species increased after the ST treatment.
