Development of a rapid and high sensitive detection system is urgently required for SARS-CoV-2 causing COVID-19. In our laboratory, biomarkers have been detected rapidly and quantitatively by grating-coupled surface plasmon-field enhanced fluorescence method using a plasmonic chip which is composed of a wavelength-order periodic structure and metal films. In this study, SARS-CoV-2 Spike protein (S-protein) was measured with a sandwich immunoassay on a plasmonic chip. Protein G was adsorbed onto the chip surface coated with thin polydopamine film. After immobilization of anti-S-protein IgG antibody, S-protein solution prepared at different concentrations was injected. As a result of detection with a fluorescently labeled S-protein antibody, S-protein was detected at a concentration of 100 pg / mL with the our detection system.
