[20a-P05-18] Formation of protein aggregates by UV laser irradiation to a cytochrome c olution

We found that insoluble aggregates of cytochrome c were formed on a glass substrate upon irradiating UV laser (λ= 405 nm). For the characterization of the aggregates, we used an amyloid sensor, namely, thioflavin T that shows bright fluorescence in the presence of amyloid fibrils. In the fluorescence image of the aggregates, they exhibited strong fluorescence derived from Thioflavin T. This result suggests that the cytochrome c aggregates have a β-sheet-rich structure like amyloid fibrils. In the presentation, we will show the pH and laser intensity dependences on the aggregate formation and discuss the mechanism.
