As the development of optogenetics, it is desired to enhance the speed of image acquisition in two-photon microscopy. The speed of image acquisition and field of view intrinsically have a trade-off relationship. In order to take as many cells as possible in a shortest time, we propose a new method to realize multiplane imaging in two-photon microscopy using diffractive optical elements, such as a spatial light modulator (SLM). By projecting appropriate phase pattern, images from different depth could focused in the different portion of the single image sensor. The proposed method is suitable for real-time measurement, because it does not require heavy post processing, and makes use of excellent axial resolution of two-photon microscopy.
