[22p-A106-8] Introduction of Biomolecules into Artificial Lipid Bilayers Using Biotin-Streptavidin Interaction

We propose to use the Biotin-Streptavidin interaction to bind vesicles to lipid bilayers and control their fusion. Vesicle fusion was triggered by addition of Ca²⁺ to the buffer solution. We found that optimizing the concentration of Streptavidin effectively promotes fusion. It increases mobile fraction in fluorescence recovery after photobleaching. The change in diffusion coefficients suggested that the fusion state progressed from hemifusion to full fusion. We will also report the results of examining the introduction of biomolecules into microwells.
