Fluorescence molecules have temperature dependence in its emission intensity, and this feature has been applied to microscopic liquid thermometry. However, the conventional intensity-based method has drawbacks of being susceptible to conditions of liquid, such as pH and liquid thickness. In this study, we focused on fluorescence anisotropy (FA) and performed temperature measurement of biochip. In this method, the influences of pH and liquid film thickness can be inherently compensated out by applying the ratiometric approach.
