[22a-P01-3] Improvement of Measurement Accuracy of DNA Microarray by Pixel Analysis of Probe Spot Fluorescent Images

This report relates to improving an accuracy of spot detection on DNA microarrays. There is a signaling array probe in which each probe modified with a fluorescent or a quenching molecule is partially bound by a complementary sequence and immobilized on a substrate in pairs. When a target hybridizes with this probe pair, quenching is canceled, and fluorescence is detected. We have analyzed a change in the amount of fluorescence in the probe spot due to hybridization in detail for each pixel of the fluorescent image. An effect of offset light due to incomplete quenching, probe immobilization amount, and reaction variation was reduced, and measurement accuracy was improved.
