[IYS-04E] Advanced in vitro evaluation system for selecting anti-diarrheal immunobiotics

[Objective] Several immunoregulatory probiotics (immunobiotics) are likely effective against intestinal infections. This study was to establish an advanced evaluation system for selecting anti-diarrheal immunobiotics for porcine host. [Methods] Porcine intestinal epithelial (PIE) cells were stimulated with 14 strains of lactic acid bacteria (LAB) individually followed by challenge with ETEC or poly (I:C), and measuring gene expressions by qPCR. Immune-related phenotypes were ranked by correlation analyses. High-ranked phenotypes were then assessed in ETEC-challenged PIE after the stimulation with 7 selected LAB strains. Host gene expressions were also compared to the genomic segments of LAB species. [Results]Immunobiotic stimulation differently affected the expression of immune related genes (phenotypes) in PIE cells. Some phenotypes had high correlations. Results suggested that not only chemical components but also physical factors of LAB genomes are important for immunobiosis.
