[IYS-07E] Investigation of factors affecting milk microbiota of dairy cows managed by automatic milking systems

Wu Haoming, Nguyen Dang Qui, Takeshi Tsuruta, Naoki Nishino (Okayama Univ.)

[Objectives] Microbiota of the milk collected manually and automatically from AMS-managed cows (Exp.1), and how the milk microbiota could interact with the cowshed environment at dairy farms managed by AMS were examined (Exp. 2).
[Methods] Samplings were made at dairy farms located in Okayama (Exp. 1 and 2) and Hiroshima (Exp. 2) prefectures. In Exp. 1, the milk microbiota was compared between May and July. In Exp. 2, feed (total mixed ration silage), rumen fluid, feces, milk, bedding, water, and airborne dust microbiota was characterized. Microbiota was assessed by Illumina MiSeq sequencing and the gene functions were predicted by PICRUSt.
[Results] The microbiota of manually collected and automatically collected milk was substantially different (Exp. 1). The three most prevalent taxa (Aerococcaceae, Staphylococcaceae, and Ruminococcaceae at Okayama farm and Staphylococcaceae, Lactobacillaceae, and Ruminococcaceae at Hiroshima farm) were shared between milk and airborne dust microbiota (Exp. 2).
