[V-16-19] Advanced development of an in-vitro IBD model for the selection of immunobiotics

[Objective] Inflammatory bowel disease (IBD) is a worldwide health problem. This study aimed to establish an advanced evaluation system using in-vitro IBD model for immunobiotics with anti-IBD.
[Methods]
Small intestinal epithelial cells were stimulated with LAB and then treated with dextran sulfate sodium (DSS) to induce inflammation. Some strains were selected by evaluating the expression of A20 and analyzing their effects on IBD-related genes, intracellular changes, and tight junctions.
[Results] In-vitro IBD model was established by stimulation with DSS. Among 20 strains, 8 strains enhanced or suppressed the expression of A20 and were selected for detailed analysis. Ca2+ flux, early inflammatory response, expression of inflammatory cytokine, and protection of tight junction protein were suppressed by A20-enhancing strains. The developed in-vitro immunoassay could be of value for the screening and selection of new immunobiotic strains for their application in IBD.
