[ODP-223] Upstream Gene Sequences of ISEcp1 in ESBL-Producing E. coli collected from Indonesia and Vietnam

Background: Recent evidence in the Southeast Asia suggest the ESBL producing-E. coli disseminated in healthy person causes public health concern. ISEcp1 has an important role to be transferred blaCTX-M which encodes CTX-M type ESBL. In this study, we compared upstream genetic structures (UGS) of ISEcp1-blaCTX-M in ESBL-producing E. coli collected from healthy person in Indonesia and Vietnam.
Method: Number of examined ESBL-producing-E. coli isolates were 196 isolates from Indonesia side and 321 from Vietnam side. Whole bacterial DNA was extracted and utilized to PCR amplification of upstream region of ISEcp1-blaCTX-M. The amplified regions were analyzed by the Nanopore sequencer.
Result: We have 318 ESBL-producing E. coli isolates containing blaCTX-M-1 genes group and 199 isolates containing blaCTX-M-9 genes group. For isolates containing blaCTX-M-1 genes group, there are 32 UGS patterns from Indonesia and 42 UGS patterns from Vietnam. Meanwhile, for isolates containing blaCTX-M-9 genes group there are 3 UGS patterns from Indonesia and 26 UGS patterns from Vietnam.
Conclusion: The results indicated there were variety of upstream genetic structures of ISEcp1-blaCTX-M in ESBL-producing E. coli isolates obtained from Indonesia and Vietnam. Further investigation is needed to explain whether that variations are related to high prevalence of ESBL-producing E. coli.
