Many studies have shown that various organelles, such as mitochondria, autophagy, and lysosomes, are involved in the regulation of neuronal morphology and functions. Among the organelles, we are focusing on the endoplasmic reticulum (ER). The ER membrane molecule Derlin-1 is involved in the pathological mechanism of amyotrophic lateral sclerosis. The CNS-specific Derl1-deficient mice show abnormalities in postweaning brain development and adult neurogenesis, resulting in impaired motor and cognitive functions. The pathological mechanisms of these mice were analyzed using fluorescence microscopy, and we also found the compounds that restore these phenotype. In this seminar, I will introduce some of these studies.
